Dynasty Warrior 6 kali ini mengenalkan inovasi baru kedalam gameplaynya. Sekarang setiap player dapat memanjat dan berenang. Bahkan ada kalanya gerakan baru tersebut menjadi bagian dari cerita. Seperti yang ada pada Battle of Fan Castle. Selain itu setiap karakter diberi tampilan serta senjata baru. Seperti misalnya Lu Bu yang kali ini tidak menggunakan Halbard sebagai senjata andalannya.
